Autophagy induced by Vip3Aa has a pro-survival role in Spodoptera frugiperda Sf9 cells

Vip3Aa is an insecticidal protein that can effectively control certain lepidopteran pests and has been used widely in biological control. However, the mechanism of action unclear. In present study, we showed could cause autophagy Sf9 cells, which was confirmed by increased numbers GFP-Atg8 puncta, appearance autophagic vacuoles, elevated Atg8-II level. Moreover, found AMPK-mTOR-ULK1 pathway involved Vip3Aa-induced autophagy, might be associated with destruction ATP homeostasis Vip3Aa-treated cells. Both p62 level GFP-RFP-Atg8 yellow fluorescent spots demonstrated cells inhibited at 24 h after treatment. With prolongation treatment time, this inhibition became more serious led to autophagosome accumulation. Genetic knockdown ATG5 or use inhibitor 3-MA further sensitivity Vip3Aa. Overexpression reduced cell mortality summary, results revealed induced a pro-survival role, related development insect resistance.
